This paper shows for the first time that estradiol rapidly activates the PI3K-dependent pathway in breast cancer MCF-7 cells. Such an activation occurs through the assembly of tripartite complex established between estradiol receptor, PI3K and Src tyrosine kinase. The assembly of this complex by estradiol ultimately triggers cyclin D1 transcription and G1/S progression in breast cancer cells.

PI3-kinase in concert with Src promotes the S-phase entry of oestradiol-stimulated MCF-7 cells.
